The BioMaster HS-qPCR Hi-ROX SYBR (2×) kit contains BioMaster HS-qPCR Hi-ROX SYBR reaction mix, 50 mM MgCl2 solution and sterile water. The mix contains all components, besides DNA template, primers and the probe, needed for PCR: HS-Taq DNA polymerase, dNTP mix, PCR buffer, Mg2+ solution, SYBR Green I, fluorescent dye ROX, and inert dye.

BioMaster HS-qPCR Lo-ROX SYBR (2×) is optimized for efficient and reproducible hot-start real-time PCR of genomic, plasmid and viral DNA samples. The mix includes additives influencing primer annealing temperature and characteristics of template melting. This results in increasing of the PCR specificity and allowing to use the templates with complicated spatial structure.

DNA polymerase included in the mix is inactive at room temperature and requires preheating at 95 °C for 5 min. Blue hue of the reaction solution provided by the inert dye allows easy control when multiwell plates are used. Composition of the kit (low concentration of Mg2+ in reaction solution and a vial with optional 50 mM MgCl2 solution) allows optimization of the reaction conditions for various primer/template systems.

The master mix is ideally suitable for PCR platforms that use ROX passive dye at high concentration as a reference guide, such as Thermo Fisher (formerly ABI)  7000, 7300, 7700, 7900, 7900HT, and StepOnePlus. Use of the kit saves time and minimizes the risk of contamination due to reduced number of pipetting steps.
