T7 RNA polymerase is a DNA-dependent RNA polymerase with strict specificity for its respective double-stranded promoters. It catalyzes the 5'→3' synthesis of RNA on either single-stranded DNA or double-stranded DNA downstream from it promoter. Incorporates modified (e.g., aminoallyl-, biotin-, fluorescein-, digoxigenin-labeled) nucleotides.
Unit definition:
One unit is defined as the amount of enzyme that will incorporate 1 nmol NTP into acid-insoluble material in a total reaction volume of 50 μl in 1 hour at 37°C.
